You can't use a Rule Province action if the province's race doesn't match the character performing the action. So a human regent with all those different subjects will need a very diverse array of lieutenants and a long, long time to achieve what you want. And you won't be able to use a lieutenant to cast pop growth to help them, either.

If this were the case, half-elves could never rule any provinces.

They count as both humans and elves for this purpose. That's how it was two games ago, at least.

Standard language, Culture, and Alignment penalties would apply. The difference can be significant when you cannot bid on the action.

It is not true, anyone can conquer/rule any race. You just have large penalties.

If you have a province on an osland and a province on the continent in the same water zone, do they count as adjacent regarding to placing holdings?

Basically for purposes of later holding placement, yes.
